Abstract: | Studying embryonic development is challenging due to the difficulty of accessing embryos in vivo and replicating their natural conditions in vitro. Traditional models like organoids and embryoid bodies often lack necessary mechanical stimuli and are limited in size. Embedded bioprinting, a 3D printing technique, offers precise control over embryonic models by allowing human embryonic stem cells to self-assemble into dense tissues. However, staining these tissues is complicated by embedding bath, and traditional staining methods damage the fragile structures. This study tests an innovative live staining technique where the staining solution is printed near the tissue, allowing it to diffuse and stain the tissue effectively. Key factors influencing staining include the concentration of the stain, the distance from the tissue, and light intensity of the microscope. The study demonstrates that higher staining concentration and reducing the distance between the staining source and the tissue resulted in brighter staining. This technique shows promise for advancing the study of early human embryonic development and tissue engineering, particularly in exploring the effect of biomechanical and biochemical factors. |
