Abstract: | Monocyte-like cell lines are invaluable as models for monocytes in research regarding natural biological states as well as in the context of pathology, treatment target discovery and drug testing. However, reports regarding findings relating monocyte-like cells are regularly contradictory due to the plasticity of monocytes and their sensitivity to culture conditions. In addition, two prominently used models, THP1 and U937, are naturally at the preceding stage of promonocytes. Hence, differentiation of the cell lines to a mature profile is required for use in research. However, maturation and differentiation of monocytes is still poorly understood, and as a result no replicable protocol for THP1 and/or U937 cell lines has yet been established. This research investigates the effect of a number of maturation protocols, and potentially influential culture specifics that are prone to vary between labs, on the maturity profile of promonocytes through flow cytometry of expression levels of CD14, CD16, CD13 and HLA-DR. |
